![]() Hakkari only gives 14 more AP, which is negigable, and the swords +6 is nice, but not enough to make it an ideal rogue set. Now I'm going to compare this w/ Dal'Rend's Arms.ĭal'Rends: You get 54 AP (4 more ap from 4 str, rogues only get 1 ap per 1 str), +7 defense +100 armor (good if you pull aggro a lot), and +1% crit. Hunters do melee in some fights, especially against the spider boss because you gotta stay close to the spider, and also because the stats on these babies give the same things it does for rogues: increased crit chance, attack power boost. Meaning if you wanted a sword or item purely for PvP, you wouldn't get it if someone else could use it for PvE. Now, technically this would be a great weapon for a fury warrior in PvP, but in my guild, since we just started doing ZG and MC, we make PvE items a priority over PvP items. I'd take Dal'Rend's over these, because honestly I lose damage in a long fight because my SS hits are MUCH lower. Sure you'll proc poison more often, or maybe get your extra swing more frequently, but the white damage on this is negigable and if you were going for that stuff a dagger with windfury would do the same white damage, probably more poison procs dual wielding daggers than this set. I can see how it would be easy to think this is a rogue set, but it's reallyl not that good for rogues. Thottbot의 댓글Ok, first off lemme say I'm a Rogue, and I use swords in PvE for the sustained DPS and daggers in PvP for the burst dps.Īnyone that says I would "roll" for this item is a retard because you don't roll in ZG, and those people should be disregarded. If i were the leader of the guild i'd make sure the hunter understand this, and if he doesnt i'd ask him politely to leave the guild. By far, this is the best 1h sword/axe available to us, so for a hunter to roll for this over a fury warrior in their guild just so he can put an extra 15agility at the cost of perhaps the best weapon for the fury warrior makes me sick to my stomach. For our mainhand we want a weapon as fast as possible to generate rage and better proc rate, but not a dagger b/c of the new normalize instant attack where daggers only get 1.7 speed of damage from AP, whereas 1h swords and axes get 2.4 speed from AP for our overpower and execute. This sword is by far, the best Mainhand weapon for an experienced fury warrior. In fact half of the best fury gears aren't even epics. The problem with fury is that a lot of ppl dont know how to spec it, what to do, and what to wear. with the right gear fury warrior can do very well in DPS compare to MS warrior and rogues. Thottbot의 댓글I'm an experienced fury warrior. Its clear that Warblade is more potent as a main hand SS weapon after patch, but by my calculations it still doesnt add up to af Krol SS attack. Lets say u have 700AP and "roll" the highest dice number on each sword: Sinister Strike still does damage according to damage range (high max sword damage = high sinister strike damage) although the speed has been fixed to 2.4 speed after patch 1.8.įor Sinister Strike I believe the formula is: Thottbot의 댓글Ill ignore the stupid 2-3 last posts and comment on some of the more intelligent further up.Īlthough speed shouldnt be taken into account with the new instant attack game mechanichs (after 1.8) it doesnt meen a low max dam, high dps sword will be good for a rogues SS damage. If your guild killed Onyxia Deathbringer would be better, but Frostbite is pretty solid and cool until you are in MC/Onyxia with your guild. Hunters and Rogues should get prio on this weapon, fury warriors should go for another weapon like (Frostbite) (Horde) Same stats but a mace if you are alliance. And of course if you can get the offhand it's a very big +++. If you are not in MC the Warblade of Hakkari is a very nice main hand. For rogues it's pretty nice but I would prefer Brutality blade in main hand because the stats are a bit better, and so is the DPS. For a fury DW warrior it is a bit slow for a Main hand, if you are a fury warrior you should try to get the off hand from the set, and (Deathbringer) that's a very nice combo imo. This is surely a great MH for a hunterĢ8 AP = +/- 28 agility for a hunter, And the 1 % crit is pretty nice as well. I got both a 60 warrior and hunter and a rogue as well. 7343의 댓글It depends on your ability's to play. ![]() ![]() ![]() Chris Rock Max Minghella and Marisol Nichols all stood out to me, giving solid performances, but the problem is that most of the dialogue is conflicts and arguments, which just wore off very fast. Saw fans will get a kick out of the traps as I said, but the movie isn't about them at all and most of them feel like they're only there for the shock value. Overall, Spiral just didn't quite do it for me when looking back on it. The look and feel of this film were absolutely one of the more impressive entries in the franchise, which is frustrating too because they deserved to be part of a better screenplay. I thought every one of his compositions were fantastic and stood out as easily the best element of the movie. What I will shine a light on though is the fact that cinematographer Jordan Oram really stepped up to the plate here. I will always appreciate when a series tries new things in order to keep it going, especially when they're only making the films for profit. It's the fact that nearly every scene is an argument or a conflict between two or more characters and it became a bit of a headache honestly. ![]() I never found myself getting attached to any of the characters either, which is a shame because they're clearly all trying here. This happened a couple of times throughout the film, so really this film is about the detective storyline with the added addition of traps to keep Saw fans amused. When the film flashes back to show whether or not the person makes it out alive or not, you're already aware so the tension was non-existent. In particular, there is a trap that occurs, but you are shown the aftermath first. On top of that, they all play out in weird ways. I never once cared about who lived and who died in these traps though, because they always seemed over-the-top in terms of how ridiculous and elaborate they were. Picking off dirty police officers who have caused people harm in ways they never realized, this killer has a very specific motive. The Jigsaw killer is officially dead, but a new killer (who is copying Jigsaw's old style) is now on the loose. It has been many years since the last installment has taken place in this world. Here's why, unless you're a massive Saw fan, you can skip Spiral. With that said, although he himself is devoted to this role and the filmmakers are clearly trying to put a new spin on the series, it just felt like more of the same, with the added layer of the detective storyline. When I heard Chris Rock had an idea and would also be starring in it, I was intrigued to see what he would do outside of his comfort zone. I love the first film for what it is and I always find myself hoping that this series will churn out something fresh and original when a new sequel is announced. I'd be lying if I didn't at least call myself a minor fan of the Saw franchise. In vivo pilus subunits function to form the main body of the helical pilus (approximately 3.28 subunits per turn), and the adhesive head. Particular subunits may have additional domains e.g. Because this N-terminal extension is the same β strand that normally completes the Ig fold, a large hydrophobic cleft is formed in the mature subunit (P1-P5), this cleft is both essential to pilus biogenesis but also necessitates a chaperone to stabilise the structure. The pilus subunit structure also has an Ig-like fold but with an essential N-terminal extension rather than a C-terminal extension. PAP pilin major protein, PapA homo13mer, top view, E.Coli. The chaperone functions to stop aggregation and degradation of pilus subunits by binding their interfacing regions and facilitating correct protein folding before bringing them to the usher/fimbriae complex. Long looped chaperones are FGL and short looped chaperones are FGS. Chaperones belong to one of two families based on the length of the loop connecting the beta strands F1 and G1. This fold is formed by 13 β strands (A1-G1) and 4 short α helices. The periplasmic chaperone (PapD) has a 'boomerang' structure formed by an immunoglobulin (Ig) like fold with an essential C-terminal extension (G1). In the active dimeric usher one of the usher has the plug located periplasmically and the other has it located inside the translocation pore. The plug domain acts to stop premature formation of a pilus and can either be located inside the pore or on the periplasmic side of the membrane. The NTD and CTDs function to bring chaperone/subunit complexes to the growing pilus and facilitate translocation across the pore. The usher pore (PapC) is formed by a 24 stranded beta barrel with 4 additional domains: N-terminal domain (NTD), Plug domain, and two C-terminal domains (CTD1 and CTD2 respectively). The usher forms the outer-membrane pore and functions in vivo as a dimer, though only one of the ushers is active at any one time. The P-pilus (pap) system is one of the best characterised and is shown below. The Pilus subunits polymerise via a non- covalent interaction to form the mature pilus which consists of an adhesive tip, helical body and an usher bound base. Overall the system includes periplasmic chaperones, periplasmic and extracellular pilus subunits, dimeric usher outer membrane pore and associated Sec machinery. Chaperone-usher secreted fimbriae are important pathogenicity factors facilitating host colonisation, localisation and biofilm formation in clinically important species such as uropathogenic Escherichia coli and Pseudomonas aeruginosa.Īll chaperone/usher systems are found within gene clusters consisting of at least an usher, a chaperone and one or more fimbriae subunits. These fimbriae are built up out of modular pilus subunits, which are transported into the periplasm in a Sec dependent manner. Chaperone-usher fimbriae (CU) are linear, unbranching, outer-membrane pili secreted by gram-negative bacteria through the chaperone-usher system rather than through type IV secretion or extracellular nucleation systems. ![]() ![]() Overall, the ECD provides a unique combination of selectivity and sensitivity. The ECD was graded A+ for limit of detection (LOD) and specificity, but it was given a C grade for qualitative speciation, linear range, and robustness, and a D grade for universal response.Ī summary of the basic principles and operation for the ECD is provided in textbooks on GC and available in ChromAcademy, LCGC North America’s online learning platform (3–5). Schug and McNair provided a summary and “grade sheet” for commonly used GC detectors (2). Interestingly, many of these early detectors, including flame ionization detectors (FIDs), thermal conductivity detectors (TCDs), and ECDs remain widely used today, along with myriad additional ionization, physical property, and spectrometric detectors. For early workers, detection was especially challenging because the detector was required to be sensitive, selective, and able to detect a low concentration or mass of analytes in a rapidly moving stream of carrier gas. The segment describes how the ECD and Lovelock’s own work provided major influences on climate science today, nearly 60 years later.ĭetection in GC remains unique in analytical chemistry because of the large number of customized detectors. He was featured in the weekly “Life Well Lived” segment, where the show features people who are not widely known to the public but made contributions to public life (1). To chromatographers, Lovelock invented the electron capture detector (ECD) for gas chromatography (GC). This summer, I was watching the news show “Sunday Today,” and heard a name familiar to many gas chromatographers: James Lovelock. We also look at the unique life and work of Lovelock, both inventor of the ECD and one of the earliest scientists to bring attention to climate change through GAIA, which explained his theory that the Earth acts as a massive living organism. In this installment, we review the ECD and its principles of operation and discuss the general tradeoffs in detection between selectivity, ease of use, and sensitivity. The ECD was crucial in the discovery of chlorofluorocarbons (CFCs) in the upper atmosphere, ultimately leading to the international agreements limiting their use and reducing the ozone hole. The ECD is among the early detectors that is used in a relatively unmodified form today. The ECD was developed in the 1960s by James Lovelock (1919–2022), who passed away earlier this year. It is highly selective and sensitive for electron-absorbing compounds, especially those containing halogens. ![]() ![]() The electron capture detector (ECD) is among the original classical detectors for gas chromatography (GC). ![]() ![]() ![]() ![]() This is a great toolbox topic to keep employees informed-not so they can fight fires, but to have the right information to ensure they don’t make fires worse or get hurt. Plus, untrained or panicked individuals typically discharge the fire extinguisher too close to the fire, causing the fire to spread. But using the wrong fire extinguisher could make the fire worse. Often when panicked, people will grab the closest fire extinguisher to put out a fire. According to the NFPA, cooking fires are the number one cause of home fires and unattended cooking is the leading cause of fires in the kitchen. The theme for this year is “Serve Up Fire Safety in the Kitchen!” This is a great topic to segway into 24/7 safety when it comes to fire prevention. Communicating these topics to employees in a safety meeting or toolbox talk will help them to be prepared for any type of emergency that they might face while at work. The National Crime Prevention Council (NCPC) designated October as Crime Prevention Month in 1984. Another good source for information to use for a toolbox talk is 5 Unexpected Ways to Reduce Noise in the Workplace. We wrote a blog post on this topic that goes beyond just handing out earplugs-check it out here: Don’t Play It by Ear When Caring for Hearing Protection. A great toolbox topic on this subject should cover how to prevent noise-induced hearing loss, a discussion about loud areas or equipment your employees’ encounter, as well as best practices for different types of hearing protection. This type of hearing loss is permanent so it’s important for workers to protect their hearing whenever possible on a job site. The graphic below illustrates how common noises can impact your hearing. Exposure to sounds 85 decibels (dB) and above can damage your hearing( and that’s the level that OSHA regulations begin). Noise-induced hearing loss happens when you’re exposed to a noise that is too loud and that lasts too long. Noise-induced hearing loss is the most common type of hearing loss-grouped under the heading sensorineural hearing loss (inner ear nerves and hair cells are damaged)-and it affects 1 in 4 people.
